Resumen
La obesidad es una de las pandemias más importantes del siglo XXI que se caracteriza por un desbalance entre la ingesta y el gasto energético, es una problemática que va en ascenso. Una de las manifestaciones de esta patología es la liberación de especies reactivas del oxígeno (ROS), mediante la alteración de la función mitocondrial del adipocito y factores de inflamación como TNF-a e IL-6, que pueden conducir a la resistencia a la insulina y la diabetes tipo II, a través de la alteración de diferentes vías de señalización como lo son las proteínas sirtrulinas, específicamente la sirtrulina 1 (SIRT1), que tiene la capacidad de inhibir la expresión de los genes controlados por el receptor nuclear PPARγ que promueve de esta manera la movilización de los ácidos grasos, desde el adipocito, y evita su almacenamiento. Otro factor implicado es el inflamasoma, el cual podría explicar el proceso inflamatorio crónico que se genera a través de la activación masiva de citoquinas pro-inflamatorias y cómo éste repercute en un factor contribuyente y generador de obesidad y diabetes mellitus mediante el daño a la célula beta del páncreas. El presente trabajo tiene como objetivo evaluar el efecto antinflamatorio del resveratrol en preadipocitos de ratón 3T3-L1 comparado con la rosiglitazona y analizar los diferentes procesos por los cuales la obesidad conduce a un proceso inflamatorio crónico comparable con enfermedades infecciosas o autoinmunes. La metodología consistió en la incubación preadipocitos de ratón de la línea celular 3T3-L1 en presencia de un cóctel de diferenciación con rosiglitazona, isobutil xantina y dexametasona, se realizaron observaciones cada dos días en un periodo de 0 a 10 días. Posteriormente se aplicaron dosis de rosiglitazona y resveratrol, y se realizaron nuevas observaciones hasta el día 6 para corroborar el efecto del resveratrol. La determinación de triglicéridos se realizó mediante una prueba cualitativa utilizando la tinción de rojo aceite y una cuantitativa mediante extracción de lípidos con 5 ml de isopropanol para luego leer la absorbancia del extracto a 510 nm en un biofotómetro Eppendorf. Una vez obtenidos los resultados se empleó el test de Anova para elaborar el análisis estadístico. Los resultados obtenidos, tanto de forma cualitativa como cuantitativa, evidenciaron que las células tratadas con resveratrol contienen menor cantidad de triglicéridos con respecto a las tratadas con rosiglitazona, debido a que esta fitoalexina estimula la actividad de la enzima sirtrulina 1 (Sirt-1), la cual inhibe al receptor nuclear PPARγ evitando la lipogénesis y la producción de factores inflamatorios por lo que puede convertirse en una alternativa terapéutica.
Abstract
Obesity is one of the most important pandemics of the XXI century, it is characterized by an imbalance between the intake and energy expenditure, and that instead of finding a solution is a problem that is on the rise. One of the manifestations of this pathology is the release of reactive oxygen species (ROS), by altering the mitochondrial function of the adipocyte and inflammatory factors such as TNF-a and IL-6, which can lead to resistance to insulin and type II diabetes, through the alteration of different signaling pathways such as sirtrulin proteins, specifically sirtrulin 1 (SIRT1), which has the ability to inhibit the expression of genes controlled by the nuclear receptor PPARγ promoting in this way the mobilization of fatty acids from the adipocyte and avoiding its storage. Another factor involved is apparently the inflammasome which could explain the chronic inflammatory process that is generated by the massive activation of pro-inflammatory cytokines and how this influences the contributing factor and generator of obesity and diabetes mellitus through damage to the beta cell of the pancreas. The purpose of our work is to evaluate the anti-inflammatory effect of resveratrol in 3T3-L1 mouse preadipocytes compared to rosiglitazone and to analyze the different processes by which obesity leads to a chronic inflammatory process comparable with infectious or autoimmune diseases. The methodology consisted in the incubation of mouse preadipocytes of the cell line 3T3-L1 in the presence of a cocktail of differentiation with Rosiglitazone, Isobutyl Xanthine and Dexamethasone, and observations were made every two days in the period between 0 and 10 days. Subsequently, doses of Rosiglitazone and Resveratrol were applied and new observations were made until day 6 to corroborate the effect of Resveratrol. The determination of triglycerides was carried out by means of a qualitative test using oil red staining and a quantitative one by means of lipid extraction with 5 ml of isopropanol and then read the absorbance of the extract at 510 nm in an Eppendorf biophotometer. Once the results were obtained, the Anova test was used to elaborate the statistical analysis. The results obtained both qualitatively and quantitatively showed that the cells treated with Resveratrol contain a lower amount of triglycerides than those treated with Rosiglitazone, this is because this phytoalexin stimulates the activity of the enzyme Sirtrulina 1 (Sirt-1), which inhibits the nuclear receptor PPARγ by preventing lipogenesis and the production of inflammatory factors, so it can become a therapeutic alternative.
